Engineered allosteric mutants of the integrin αMβ2 I domain: structural and functional studies

چکیده
The α-I domain, found in the α-subunit of the leucocyte integrins such as αMβ2 and αLβ2, switches between the open and closed tertiary conformations, reflecting the highand low-affinity ligand-binding states of the integrin that are required for regulated cell adhesion and migration. In the present study we show, by using point mutations and engineered disulphide bonds, that ligand affinity can be reduced or increased allosterically by altering the equilibrium between the closed and open states. We determined equilibrium constants for the binding of two ligands, fibrinogen and intercellular cell-adhesion molecule 1, to the αM-I domain by surface plasmon resonance, and determined crystal structures of a low-affinity mutant. Locking the domain in the open conformation increases affinity by a factor of no greater than 10, consistent with a closely balanced equilibrium between the two conformations in the absence of ligand. This behaviour contrasts with that of the unliganded αL-I domain, for which the equilibrium lies strongly in favour of the closed conformation. These results suggest significant differences in the way the parent integrins regulate I domain conformation and hence ligand affinity.

Complex Structure of Engineered Modular Domains Defining Molecular Interaction between ICAM-1 and Integrin LFA-1
Intermolecular contacts between integrin LFA-1 (α(L)β(2)) and ICAM-1 derive solely from the integrin α(L) I domain and the first domain (D1) of ICAM-1. This study presents a crystal structure of the engineered complex of the α(L) I domain and ICAM-1 D1. Previously, we engineered the I domain for high affinity by point mutations that were identified by a directed evolution approach. In order to ...
متن کاملStructure and allosteric regulation of the X 2 integrin I domain
The integrin X 2 (CD11c CD18, p150,95) binds ligands through the I domain of the X subunit. Ligands include the complement factor fragment iC3b, a key component in the innate immune defense, which, together with the expression of X 2 on dendritic cells and on other leukocytes, suggests a role in the immune response. We now report the structure of the X I domain resolved at 1.65 Å by x-ray cryst...
متن کاملTherapeutic antagonists and the conformational regulation of the beta2 integrins.
The beta2 integrins are validated therapeutic targets for inflammatory disorders. Two distinct mechanistic classes of small molecule inhibitors, termed alpha I allosteric and alpha/beta I-like allosteric antagonist, have recently been developed. The alpha I allosteric antagonists bind underneath the C-terminal helix of the I domain and stabilize the I domain in the inactive closed conformation....
متن کاملDirected Improvement of i-Photina Bioluminescence Properties, an Efficient Calcium-Regulated Photoprotein
Photoproteins are excellent reporter systems because they don’t have virtually background signal. Aequorin is the most well-known photoprotein. Three improved engineered photoproteins photina, i-photina and c-photina, were also recently developed and optimized for generation of Ca2+ mobilization assays precisely. The total light emission is greater than aequorin and their reacti...
متن کاملRationally designed integrin beta3 mutants stabilized in the high affinity conformation.
Integrins are important cell surface receptors that transmit bidirectional signals across the membrane. It has been shown that a conformational change of the integrin beta-subunit headpiece (i.e. the beta I domain and the hybrid domain) plays a critical role in regulating integrin ligand binding affinity and function. Previous studies have used coarse methods (a glycan wedge, mutations in trans...
